When sighting in a rifle. Is it going to shoot differently at 100 yards after several shots and the barrel is hot? Obviously we hunt with cold barrels so is it best to shoot a couple of rounds, make adjustments and wait a few minutes then shoot another couple?

Yes, rifle accuracy is typically affected by a hot barrel. Letting the barrel cool between shots when sighting in a scope is recommended.

A few minutes if you have the time. I have found that I can make a few shots with a typical rifle before the barrel gets noticeably hot and I start getting flyers. If I have the time I will wait around 5 minutes between shots when sighting in. Also depends on outdoor temperature. I use a cooling rod for my muzzleloader. There are videos on YouTube about sighting in a scope and "breaking in" a new rifle barrel.

Yes. A cold bore shot is always the most accurate shot. There are many variables that affect shooting and accuracy.
